Title: Water pump to rear coolant rail interface

Simple question - does the o-ring suffice or should one also use a gasket? If it's the latter I'll probably have to resort to making one, as I don't appear to have one in my full engine set...

I made a gasket. I was not prepared to trust the o ring alone.

It’s worth checking the flatness of the flange. They can bow a bit hence the leaks. I always run a flat stone over the flange to true it up.

I was wondering all this.  I did find what looks the same but a gasket on other twin-cam models

I will check for flatness also.  do I use the o-ring and gasket, or is that silly?  Do I use other products like wellseal on the gasket?

The Delta is a different pump and symmetrical gasket - you need the stock Montecarlo gasket which is offset and never in the kits:

The gasket works well on the monte with a spray coat of blue Hylomar, it didn't have the O-ring on its flange  (y)

Mine is holding up with just the O ring and a generous
dollop of blue Hylomar. I may have used a slightly fatter ring.

Mark, if this is a new after-market rail, check that the flange is orientated
correctly in relation to the mount at the other end.

I am going to use the original I am pretty sure OEM bottom rail as the stainless one is not quite the right dims.  Top aftermarket stainless steel one looks good and has the mod for the coolant in and out from the back of the head.  I make a call on o-ring or gasket later this week.  It had a o-ring one fitted and wasnt leaking.  Mind you I now have a new pump fitted so that will sure make it leak  :P
